Horace Holly is a Cambridge professor who becomes the guardian of his late friend's son, Leo Vincey, after promising to care for him until adulthood. When Leo turns twenty-five, they open a locked box containing an ancient message that reveals a family quest to avenge an ancestor and uncover the mystery of an immortal queen known as Ayesha, or "She-who-must-be-obeyed." Holly, Leo, and their servant Job travel to Africa, where they encounter dangerous tribes, ancient ruins, and the beautiful yet powerful Ayesha. She believes Leo is the reincarnation of her long-dead lover, Kallikrates, and kills Leo's companion, Ustane, out of jealousy. Although Holly recognizes Ayesha's cruelty, he is captivated by her intelligence and supernatural abilities. In the end, Ayesha attempts to prove her immortality by entering the mystical Pillar of Fire again, but instead rapidly ages and dies, promising that she will one day return. Holly and Leo escape the lost city of Kôr and return to Cambridge, forever changed by their extraordinary adventure.
